Structure for Gold Conjugates
Aurosphere
Gold contains a constant 62.5 milligrams of gold per liter of
suspension, regardless of the size of the particles. Therefore, changing
the diameter of the particles changes the number of particles per liter.
The concentration of gold in the suspension is approximately correlated
with the absorbance (optical density) of the plasmon band in the visible
region of the electromagnetic spectrum. For Aurosphere Gold, the
OD of the "bare" particles is about 1.4 to 1.6 at their absorption
maximum of 520 to 530 nm.
Colloidal
gold particles are stabilized and converted into immunoreagents by coating
them with various proteins. The
amount of product is quantified by measurement of the optical density
(OD) of the conjugate at its absorption maximum (lambda max) in the
visible region, as with bare particles. After passive or covalent conjugation
of the protein to the gold particles, the conjugate can be concentrated
and purified by centrifugation or other techniques. The resulting material
may be quite concentrated, having an OD in the hundreds. For the purposes
of pricing, the volume of the product at its measured OD is extrapolated
to that volume which would have an OD of 10. Prices are then quoted
per milliliter of conjugate at OD 10. The cost per millileter is scaled
according to the total volume of the preparation or lot.
For example,
a conjugate is prepared with a volume of 5 ml and an OD of 70. If this
suspension were diluted to 35ml it would have an OD of 10. The price
would be determined for 35ml of product.
